Growth response on Vanda Sanderiana (Reichb. f.) to in vitro culture using various vessels and closures / Sharon M. Quezon

By: Material type: TextTextLanguage: English Publication details: 2003Description: 26 leavesSubject(s): Dissertation note: Thesis (BS Biology) -- University of the Philippines Mindanao, 2003 Summary: Three types of each culture vessel and closure were used in the observation of the growth response of V. sanderiana to in vitro culture. The vessels were 310-ml ketchup bottle, 210-ml mayonnaise bottle and 950-ml mayonnaise bottle. The closures were polypropylene, aluminum foil and cheesecloth. Results show that the difference in growth and survival in polypropylene closure aluminum foil was not significant at 5%level. However, both polypropylene and aluminum were significantly better than cheesecloth closure. Aluminum foil, like polypropylene closure, was effective in retarding moisture loss while the use of cheesecloth closure led to early moisture loss and eventually desiccation of the medium. Result on the type of vessel showed that the 10-ml ketchup bottles gave a significantly higher survival than the 950-ml mayonnaise bottles. However, survival in the 310-ml ketchup bottle was not significantly different from that obtained with the 210-mlmayonnaise bottle. In addition, a higher contamination rate was recorded from the 950-ml mayonnaise bottle compared to the 310-ml ketchup bottle due to its larger opening. The use of 310-ml ketchup bottle is therefore recommended using either polypropylene closure or aluminum foil closure, whichever is more economical or available
