520 _aSeveral studies have shown that phenotypic variations in sago palm such as armature (presence or absence of spines) are not associated with genetic variation. It is hypothesized that epigentic modification such as DNA methylation might be correlated with armature in sago palm. This hypothesis was investigated in this study with the aims to determine the biological role of DNA methylation and to verify if the relationship between transcription and DNA methylation profiles of sago palm through HPLC analysis. Thus included optimization of HPLC conditions for better resolution of peaks which further aid in detecting the peaks of dC and 5mdC in the DNA samples analyzed. Results showed that the wavelength of 280 nm used to monitor effluents efficiently distinguished signal from noise, while flow rate 0.2 mL/min resulted to peaks more distinguishable form each other. HPLC analysis of DNA methylation showed 5mdC was 51.9084% for spiny juvenile while 51.917% for non-spiny juvenile sago palms.
