Membrane solubilization of porcine pulmonary and renal angiotensin I-converting enzyme (ACE; E.C. 3.4.15.1) / Ara Mae Magno Tirol

The tissues from lung and kidney were solubilized using four different procedures after which the activities were measured by spectrophotometric method in the presence of quinolone and benzenesulfonyl chloride (BSC). The initial ACE activity from lung and kidney extract was 0.0409 U/mg and 0.0788 U/mg, respectively. The first method was by incubation with shaking at room temperature which resulted to an activity of 0.0393 U/mg for lungs and 0.1008 U/mg for kidney. The second procedure which used sonication to solubilize the membranes resulted to activities of 0.0390 U/mg for lungs and 0.0828 U/mg for kidneys. The third procedure was the addition of EDTA with the presence of Triton X-100 which decreased the activities to ? 0.0019 U/mg for lungs and 0.0015 U/mg for kidneys. The last procedure was the addition of the detergent using Triton X-100. The activity was reported to have increased to 0.1639 U/mg for lung and 0.3374 U/mg for kidney ACE. From the results of the four solubilization procedures the best procedure was the employment of Triton X- 100 which significantly increased the activities for both lung and kidney ACE
